Effects of Different Types of Helicobacter Pylori on the GJIC Function in GES-1 Cells

ABSTRACT
Objective
To study the effects of different types of H.pylori on the gap junction intercellular
communication (GJIC) function in GES-1 cells. To investigate the types of H.pylori those are related to the dysfunction of GJIC.
Methods
Different types of H.pylori clinical strains were isolated and cultured, including the Eastern-Asian type CagA+H.pylori, the Western type CagA+H.pylori, and the CagA-H.pylori. We co-cultured these H.pylori strains with GES-1 cells for 24 hours and 48 hours, respectively. The control groups were cultured without any H.pylori
for 24 hours and 48 hours. The change of the GJIC function in GES-1 cells was detected by the scrape-loading dye transfer technique (SLDT). The cell proliferation of each
group was examined by the Methyl Tetrazolium (MTT) method.
Results
The control group showed higher GJIC function in GES-1 cells, and the fluorescent dye migrated 4-5 rows to the adjacent cells at 24 and 48 hours. The GJIC function of CagA-H.pylori group decreased compared with the control group, and the fluorescent
dye migrated 3 rows to the adjacent cells. The GJIC function of the Western
type CagA+H.pylori group decreased compared with the control group and CagA-H.pylori, and the fluorescent dye migrated 1-2 rows to the adjacent cell. The Eastern-Asian type CagA+H.pylori group had no GJIC function or lower GJIC function,
the most of fluorescent dye was confined to the area of scratched single row cells and
only a few migrated 1-2 rows to the adjacent cell. The difference of cell proliferation between CagA-H.pylori group and the control group had no statistical significance. The cell proliferation
of the Western type CagA+H.pylori group and Eastern-Asian type CagA+H.pylori group at bacteria/cells ratio of 100: 1 and 200:1 were higher
than that of the control group. However, the cell proliferation of the Eastern-Asian type CagA+H.pylori group at bacteria/cells ratio of 400: 1 was significantly
lower than that of the control group for 48 hours.
Conclusions
The H.pylori can inhibit the GJIC function in GES-1 cells. The effect was associated with the CagA+H.pylori, especially with Eastern-Asian type CagA+H.pylori. The effect of H.pylori on the proliferation of GES-1 cells was associated with virulence factor CagA.
